黄芪、当归对鼠慢性肾脏纤维化的影响及相关基因的分离

摘要

To isolate genes associated with the protective effects of astragalus, angelicas on renal fibrosis in rats, chronic renal fibrosis was induced by repeated peritoneal injections of puromycin in SD rats. Rats were randomly divided into normal control (n=7),NS control (n=7) and A - A (3 mL/d,n=7) treated groups for 12 weeks. The silver staining mRNA differential display (DD) was used to investigate changes of gene expression in kidneys of the 3 groups. It is resulted that twenty-six candidate cDNA fragments were isolated by DD. Northern blot analysis of eight randomly chosen candidate cDNA fragments demonstrated one exhibiting reduced mRNA expression in the fibrogenic process and upregulated by A - A treatment. The gene is also expressed in lung, heart, liver and spleen. Homology analysis through BLAST revealed that this gene was a novel one. So, the downregulated expression of the isolated new gene may contribute to the renal fibrogenic process and be associated with A - A's protective effects.
