摘要

The objective is to develop an HPLC-MS/MS method for determining the concentration of DPTM in mouse plasma,and make a foundation for its pharmacokinetic study[1].HPLC and Mass spectrometry:Calibration standards and bio-sampleswere treated with protein precipitation method by acetonitrile.Before analysis,the residue was dissolved with certain volume mobile phase and filtered through 0.45m membrane,then separated with a ZORBAX SB-C 18 column(4.6 mm250 mm,5m)column at the flow rate of 0.2 mL/min by isocratic elution.The temperature of column oven was set at 40℃.The mobile phase consisted of acetonitrile-water with 0.1%formic acid(50:50,v/v).The auto-sampler was conditioned at 6℃and the injection volume was 10L for analysis.
