Mobilization of Autologous Peripheral Blood Stem Cells Using Mitoxantrone-Based Combinative Regimens and Harvesting by Large- Volume Leukapheresis

摘要

Objective To observe the efficacy of mitoxantrone - based combinative chemotherapy with the combination of granulocyte colony - stimulating factor (G- CFS ) and granulocyte - macrophage colony - stimulating factor( GM - CFS) on autologous peripheral blood stem cells mobilization. Methods each, subcutaneously injected from the day of white blood cell(WBC) recovery from nadir to the day before the end of APBSC harvesting. APBSC harvesting started when WBC recovered to ( 8.02 ± 3.65) ×109/L and CD34 + cells percentage ( 1.68 ± 0.79) % and finished when accumulated mononuclear cells ( MNC ) ＞ 5 × 108/kg. CFU - GM assay and CD34 + cells detection of the APBSC were performed. Results Twenty cases underwent the APBSC mobilization. APBSC harvest began at day 19.30 ± 2.12 for two successive days. Accumulated MNC was (4.67 ± 2.11) × 108/kg, CD34+ cells(7.86 ± 2.14) × 106/kg and CFU - GM(2.86 ± 2.10) × 105/kg. No severe toxicity was observed. Hematopoietic reconstitution was very well in 18 patients receiving the APBSC transplantation. Conclusions Mitoxantronebased chemotherapy combined with G - CFS plus GM - CFS was a safe and highly effective method for APBSC mobilization.
